Simultaneous Inhibition of Heat Shock Response and Autophagy with Bimetallic Mesoporous Nanoparticles to Enhance Mild‐Temperature Photothermal Therapy

Mild-temperature photothermal therapy (MPTT) is a promising tumor therapeutic modality because it can avoid the damage of normal tissues near caused by excessive heat. However, its effect severely impaired cells develop heat resistance and self-repair activating shock response cell autophagy. Herein, tannic acid–iron ion metal organic framework-coated, chloroquine (CQ)-loaded mesoporous PdPt nanosystem ([email protected]) developed to enhance MPTT simultaneous suppression [email protected] exhibits good peroxidase (POD)-mimic activity performance. As result, reactive oxygen species generated POD-mediated decomposition endogenous hydrogen peroxide mitochondria, leading limitation adenosine triphosphate supply, which suppresses upregulation proteins during MPTT, making more sensitive stress. Concurrently, CQ released from inhibits autophagy, thereby interrupting pathway cells. Consequently, significantly enhances effect, effectively inhibiting progression in 4T1 tumor-bearing mice.
